I just had a similar problem to yours. My car sat all weekend as I was visiting the folks. When I got on the expressway to drive home, the torque converter would not lock. I did not get it to lock for at least 10-15 miles and it only did so after I put it in neutral, revved it a few times, and put it back in 'D'. I was driving in '3' until I hit the highway.
I wouldn't worry about replacing the tranny just yet. It may have just been a fluke and the fluid didn't warm up fast enough to drop into OD. Have the fluid run through the radiator does help with cold starts.
